Biological Background: ELAVL2 Function and Localization
- ELAVL2, also known as HuB, HEL-N1, and nervous system-specific RNA-binding protein Hel-N1, is encoded by the ELAVL2 gene in humans.
- It is an RNA-binding protein that specifically interacts with the 3' untranslated region (3'UTR) of target mRNAs, recognizing a GAAA motif.
- ELAVL2 can bind to its own 3'UTR, as well as the 3'UTRs of FOS and ID transcripts, suggesting regulatory roles in mRNA stability or translation.
- Expression of ELAVL2 is restricted to brain and neural tissues, underscoring its specialized function in the nervous system.
- The protein undergoes alternative splicing and phosphorylation, as indicated by UniProt keywords, which may modulate its activity or interactions.
- Predicted molecular weight is approximately 39.5 kDa.
Experimental Guidance and Technical Tips
- The immunogen is an N-terminal peptide (aa 1-100); thus, the antibody is expected to recognize the N-terminal region of ELAVL2 and may detect isoforms containing this sequence.
- For Western blot (WB), a specific band at approximately 40 kDa is anticipated; neural tissue lysates (e.g., brain) are recommended as positive controls given the neural-specific expression.
- For immunohistochemistry (IHC-P), paraffin-embedded brain or neural tissue sections can be tested; appropriate antigen retrieval and antibody dilution should be empirically determined.
- ELISA can be performed using the immunizing peptide as a coating antigen; cross-reactivity with mouse and rat ELAVL2 supports multi-species assay development.
- As with any polyclonal antibody, lot-to-lot variation is possible; validation in the intended application is advised.
