Biological Background: EIF5B Function and Localization
- EIF5B (eukaryotic translation initiation factor 5B), also known as IF2, is a ~139 kDa GTP-binding protein belonging to the translation initiation factor family.
- It acts as a ribosome-dependent GTPase, promoting the joining of the 60S ribosomal subunit to the pre-initiation complex to form the 80S initiation complex. Related references: PMID:10659855 PMID:35732735
- Together with eIF1A (EIF1AX), it orients the initiator methionine-tRNA in a conformation that permits 60S subunit joining. Related references: PMID:12569173 PMID:35732735
- GTP hydrolysis is essential for eIF1A ejection and subsequent EIF5B release, enabling the formation of elongation-competent ribosomes. Related references: PMID:10659855 PMID:35732735
- EIF5B is localized in the cytoplasm.
- It is a phosphoprotein and metal-binding protein with critical roles in protein biosynthesis.
- In contrast to its prokaryotic homolog, EIF5B does not promote recruitment of Met-tRNA to the small ribosomal subunit. Related references: PMID:10659855
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–273) of human EIF5B. Consider that the antibody may detect full-length EIF5B and possibly N-terminal fragments or isoforms.
- For Western blotting (WB), use appropriate positive controls (e.g., HeLa, HEK293, or other cell lines expressing EIF5B) and validate band specificity with molecular weight markers (expected ~139 kDa). Adjust primary antibody dilution based on initial titration experiments.
- In immunofluorescence/immunocytochemistry (IF/ICC), include permeabilization steps and consider co-staining with organelle markers to confirm cytoplasmic localization.
- For ELISA, optimize coating antigen and detection conditions; the antibody may be used as a capture or detection reagent depending on the assay format.
