Biological Background: eIF5A Function and Localization

  • eIF5A (eukaryotic translation initiation factor 5A-1) is a translation factor that promotes translation elongation and termination, particularly upon ribosome stalling at specific amino acid sequence contexts (PubMed:33547280). It binds between the E and P sites of the ribosome and functions in rescuing stalled ribosomes, being specifically required for efficient translation of polyproline-containing peptides (By similarity). Related references: PMID:33547280
  • Acts as a ribosome quality control (RQC) cofactor by joining the RQC complex to facilitate peptidyl transfer during the CAT tailing step (By similarity).
  • Involved in actin dynamics, cell cycle progression, mRNA decay, and stress response pathways, including maintenance of cell wall integrity (PubMed:16987817). Related references: PMID:16987817
  • With syntenin SDCBP, functions as a regulator of p53/TP53 and p53/TP53-dependent apoptosis (PubMed:15371445). Also regulates TNF-mediated apoptosis (PubMed:15452064, PubMed:17187778). Related references: PMID:15371445 PMID:15452064 PMID:17187778
  • Mediates effects of polyamines on neuronal process extension and survival (PubMed:17360499). Related references: PMID:17360499
  • Required for autophagy by assisting the ribosome in translating ATG3 at a specific 'ASP-ASP-Gly' motif, thereby increasing ATG3 translation efficiency and facilitating LC3B lipidation and autophagosome formation (PubMed:29712776). Related references: PMID:29712776
  • Cellular cofactor of human T-cell leukemia virus type I (HTLV-1) Rex protein and human immunodeficiency virus type 1 (HIV-1) Rev protein, essential for mRNA export of retroviral transcripts.
  • Subcellularly localized to the cytoplasm, nucleus, and endoplasmic reticulum membrane. Expressed in umbilical vein endothelial cells and various cancer cell lines.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide covering N-terminal residues 1-100 of human eIF5A. This region is relatively conserved; cross-reactivity with mouse and rat may be expected, but validation in the relevant sample system is recommended.
  • For Western blot (WB), eIF5A has a predicted molecular weight of ~17 kDa; ensure appropriate gel percentage and transfer conditions for low-molecular-weight proteins.
  • For ELISA, consider using the antibody as a capture or detection reagent; optimal dilutions should be determined empirically.
  • As eIF5A undergoes hypusine modification, the antibody may recognize both unmodified and modified forms. Verify specificity if studying post-translational modifications.
  • eIF5A is expressed in many cancer cell lines; select an appropriate positive control when performing assays.
