Biological Background: Eukaryotic Translation Initiation Factor 5 (eIF5) Function and Localization
- Eukaryotic translation initiation factor 5 (eIF5) is encoded by the EIF5 gene and is a key component of the 43S pre-initiation complex (43S PIC) that scans the mRNA 5' untranslated region to locate the start codon. Related references: PMID:11166181, PMID:22813744, PMID:24319994
- Within the 43S PIC, eIF5 acts as a GTPase-activating protein (GAP) that stimulates GTP hydrolysis by the eIF2 gamma subunit (EIF2S3). Related references: PMID:11166181
- During scanning, the N-terminal domain (NTD) of eIF5 interacts with EIF1A, while its C-terminal domain (CTD) binds EIF1, helping maintain EIF1 in the open PIC conformation. Related references: PMID:22813744, PMID:24319994
