Biological Background: Eukaryotic Translation Initiation Factor 5 (eIF5) Function and Localization

  • Eukaryotic translation initiation factor 5 (eIF5) is encoded by the EIF5 gene and is a key component of the 43S pre-initiation complex (43S PIC) that scans the mRNA 5' untranslated region to locate the start codon. Related references: PMID:11166181, PMID:22813744, PMID:24319994
  • Within the 43S PIC, eIF5 acts as a GTPase-activating protein (GAP) that stimulates GTP hydrolysis by the eIF2 gamma subunit (EIF2S3). Related references: PMID:11166181
  • During scanning, the N-terminal domain (NTD) of eIF5 interacts with EIF1A, while its C-terminal domain (CTD) binds EIF1, helping maintain EIF1 in the open PIC conformation. Related references: PMID:22813744, PMID:24319994
  • Upon start codon recognition, eIF5 NTD triggers eIF2γ GTP hydrolysis, inducing a closed PIC state that increases CTD affinity for EIF2β, releases EIF1, and stabilizes the closed complex. Related references: PMID:11166181, PMID:22813744
  • eIF5 localizes predominantly to the cytoplasm where translation initiation occurs.
  • The protein has a theoretical molecular weight of approximately 49 kDa and is subject to post-translational modifications including phosphorylation and ubiquitination.

Experimental Guidance and Technical Tips

  • For western blotting (WB), consider using a total protein loading control and optimizing primary antibody dilution based on the approximately 49 kDa target band.
  • When performing immunofluorescence (IF/ICC), include appropriate permeabilization and blocking steps; validate antibody specificity in the relevant species (human, mouse, rat).
  • For ELISA, use the recombinant EIF5 immunogen (amino acids 1-235) as a positive control to generate a standard curve.
  • The antibody is raised against full-length human EIF5 N-terminal region (aa 1-235); confirm reactivity in your sample system and consider cross-reactivity with other eIF5 family members.
