Biological Background: eIF4E Function and Localization
- Eukaryotic translation initiation factor 4E (eIF4E) is also known as eIF-4F 25 kDa subunit or mRNA cap-binding protein, encoded by the EIF4E gene. It is a central regulator of protein synthesis.
- Recognizes and binds the 7-methylguanosine (m7G) cap at the 5' end of mRNAs as a core component of the eIF4F complex, facilitating unwinding of mRNA secondary structures and ribosome recruitment for translation initiation. Related references: PMID:16271312 PMID:22578813
- Mediates nuclear export of a subset of mRNAs, including cyclin D1 (CCND1), NOS2, and MDM2, through recognition of the 4ESE element and assembly of a transport complex involving LRPPRC and XPO1. Related references: PMID:23471078 PMID:22684010
- Functions in translational repression as part of the CYFIP1-EIF4E-FMR1 complex and participates in the storage of translationally inactive mRNAs in cytoplasmic processing bodies (P-bodies). Related references: PMID:24335285

Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human eIF4E protein (amino acids 1–217). This antibody is expected to detect endogenous eIF4E at approximately 25 kDa in human samples.
- For Western blot (WB), consider using whole-cell lysates from HeLa, HEK293, or Jurkat cells as positive controls. A dilution range of 1:500–1:2000 is common for polyclonal antibodies, but optimization is recommended.
- In immunofluorescence/immunocytochemistry (IF/ICC), eIF4E localizes mainly to the cytoplasm, with nuclear and P-body staining possible depending on cell type and conditions; validate staining patterns in your specific system.
- For ELISA applications, this antibody can be used as a capture or detection reagent; titrate to determine the optimal working concentration for your assay format.
