Biological Background: eIF4A3 Function and Localization
- eIF4A3 is an ATP-dependent RNA helicase belonging to the DEAD-box family, essential for ATP-dependent unwinding of RNA secondary structures. Related references: PMID:16170325
- It is a core component of the exon junction complex (EJC), deposited 20–24 nucleotides upstream of exon–exon junctions during pre-mRNA splicing. The EJC influences mRNA export, localization, translation, and nonsense-mediated decay. Related references: PMID:16170325, PMID:16209946, PMID:16314458
- eIF4A3 participates directly in the spliceosome and pre-mRNA splicing machinery. Related references: PMID:11991638, PMID:22961380, PMID:28076346
- It modulates alternative splicing of BCL2L1/Bcl-X, inhibiting the production of pro-apoptotic Bcl-X(S) isoforms, thus affecting apoptosis. Related references: PMID:22203037
- The protein is required for normal craniofacial development. Related references: PMID:24360810
- Subcellularly, eIF4A3 localizes mainly to the nucleus and nuclear speckles, but can shuttle to the cytoplasm as part of the EJC on mature mRNAs.
- Post-translational modifications include phosphorylation, acetylation, and ubiquitination (isopeptide bond formation), as indicated by UniProt keywords. Alternative names include DDX48, NMP265, eIF4AIII, and KIAA0111; the protein is ubiquitously expressed.
Experimental Guidance and Technical Tips
- The immunogen comprises the N-terminal 210 amino acids of human eIF4A3; antibody reactivity is expected to detect endogenous full-length protein in human, mouse, and rat samples.
- For Western blotting, the predicted molecular weight is approximately 47 kDa; consider using nuclear extracts or whole-cell lysates from proliferating cells, as eIF4A3 is predominantly nuclear and associates with spliceosomes.
- In ELISA applications, the antibody can be used with recombinant eIF4A3 protein or peptide coating; validate assay conditions with appropriate positive and negative controls.
- Verify antibody specificity in your experimental system by assessing knockout or knockdown controls where available.
