Biological Background: eIF4A Function and Localization
- eIF4A-1 (P60842), also known as DDX2A, is an ATP-dependent RNA helicase and a core subunit of the eIF4F translation initiation complex.
- As part of eIF4F, eIF4A-1 participates in cap-dependent translation initiation by unwinding RNA secondary structures in the 5'-UTR of mRNAs, enabling ribosome binding and scanning for the start codon.
Related references: PMID:20156963 - This helicase activity is essential for efficient protein synthesis and contributes to cell proliferation and growth.
- The eIF4A family consists of three highly homologous members: eIF4A-1 (P60842), eIF4A-2 (Q14240), and eIF4A-3 (P38919), all sharing conserved DEAD-box helicase domains.
- Subcellular localization includes the cytoplasm, perinuclear region, cell membrane, and stress granules, where it is recruited under cellular stress.
- Post-translational modifications such as acetylation, phosphorylation, and ubiquitination regulate its activity and stability.
- eIF4A-1 is also implicated in host-virus interactions, playing roles in the replication of certain viruses.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering the N-terminal region (aa 1-100) of human eIF4A-1. The resulting polyclonal antibody may recognize multiple eIF4A family members due to high sequence conservation; consider verifying specificity for your target isoform in the relevant cell or tissue lysate.
- For Western blotting, a dominant band around 46 kDa is expected for eIF4A-1, with possible additional bands corresponding to eIF4A-2 (approx. 46 kDa) and eIF4A-3 (approx. 48 kDa). Use appropriate positive and negative controls.
- In immunofluorescence/ICC, staining may be observed in the cytoplasm and nucleus, with possible enrichment at the perinuclear region or in stress granules under stress conditions. Pre-treatment of cells to induce stress granule formation (e.g., with arsenite) can be used for localization studies.
- For ELISA, the antibody is suitable as a capture or detection reagent in sandwich formats; cross-reactivity with other eIF4A family proteins should be assessed empirically.
- As a polyclonal antibody, consistent performance across lots should be validated; consider employing internal reference standards for quantitative assays.
