Biological Background: EIF3M Function and Localization

  • EIF3M (eukaryotic translation initiation factor 3 subunit M) is also known as fetal lung protein B5 (hFL-B5), PCI domain-containing protein 1 (PCID1), B5, GA17, TANGO7, and hfl-B5. It is encoded by the EIF3M gene (also designated HFLB5, PCID1) and serves as a subunit of the eIF-3 complex.
  • EIF3M is an essential component of the eIF-3 complex, which is required for multiple steps in translation initiation: the complex binds the 40S ribosome, recruits initiation factors to assemble the 43S pre-initiation complex, stimulates mRNA scanning, and promotes disassembly of post-termination ribosomes. Related references: PMID:17403899, PMID:25849773, PMID:27462815
  • Through differential RNA stem-loop binding, the eIF-3 complex (via EIF3M) exerts translational activation or repression on a subset of mRNAs controlling cell proliferation, differentiation, and apoptosis. Related references: PMID:25849773
  • EIF3M is predominantly localized in the cytoplasm, consistent with its function in translation initiation.
  • The protein is broadly expressed across human tissues.
  • Post-translational modifications include acetylation and phosphorylation; alternative splicing generates multiple isoforms. Additionally, EIF3M may play a role in viral entry, as it has been implicated in facilitating infection by herpes simplex virus 1 (HSV-1) and herpes simplex virus 2 (HSV-2).

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant protein covering the full-length EIF3M (amino acids 1–374). This design may react with all isoforms; nevertheless, it is advisable to validate the antibody in your specific sample system.
  • For Western blot (WB), the predicted molecular weight is ~42 kDa. Use appropriate loading controls and run a positive control lysate from human, mouse, or rat cells, as the antibody shows cross-reactivity with these species.
  • For immunohistochemistry (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 9.0) should be optimized empirically. Test on known positive tissues such as proliferative tumor sections, given EIF3M's role in cell cycling.
  • In ELISA applications, the polyclonal nature may provide robust capture; consider pairing with a monoclonal anti-EIF3M antibody for a sandwich ELISA format if quantification is required.
  • Because EIF3M is part of a large multi‑protein complex, co‑immunoprecipitation experiments may be feasible, but confirm complex integrity under your lysis conditions.
