Biological Background: EIF3G Function and Localization
- EIF3G (eukaryotic translation initiation factor 3 subunit G) is also known as EIF3S4, eIF3-p44, and eIF3-delta. It is a core subunit of the eIF-3 complex, which plays a critical role in the initiation of protein synthesis by associating with the 40S ribosomal subunit and facilitating the recruitment of initiation factors to form the 43S pre-initiation complex. Related references: PMID:17581632
- The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition, and is required for disassembly and recycling of post-termination ribosomal complexes, preventing premature 40S–60S joining. Related references: PMID:17581632
- EIF3G specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation, and apoptosis, using different modes of RNA stem-loop binding to exert either translational activation or repression. Related references: PMID:25849773
- This subunit can bind 18S rRNA, suggesting a direct interaction with the ribosomal small subunit.
- In cases of feline calicivirus (FCV) infection, EIF3G plays a role in the ribosomal termination-reinitiation event leading to the translation of VP2. Related references: PMID:18056426
- Subcellular localization includes the cytoplasm, nucleus, and perinuclear region, consistent with its roles in translation initiation and potential nuclear functions.
- Post-translational modifications include phosphorylation, and the protein is identified as a phosphoprotein and RNA-binding protein by proteomics studies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human EIF3G (amino acids 1-320), which may recognize multiple isoforms or degradation products. Validate specificity in your experimental system using appropriate controls.
- For Western blot, a major band at approximately 36 kDa is expected, but additional bands may arise due to post-translational modifications or association with other eIF-3 subunits.
- The antibody is suitable for immunofluorescence/immunocytochemistry; consider optimizing fixation and permeabilization conditions for your cell type.
- When performing ELISA, titrate the antibody to determine the optimal working concentration for your assay format.
