Biological Background: eIF3e Function and Localization
- Eukaryotic translation initiation factor 3 subunit E (eIF3e), also known as INT6, EIF3S6, or eIF3-p48, is encoded by the EIF3E gene (Gene ID: 3646).
- Component of the eIF-3 complex required for several steps in protein synthesis initiation, including 43S pre-initiation complex formation, mRNA recruitment, ribosomal subunit recycling, and prevention of premature 40S-60S joining. Related references: PMID:17581632, PMID:25849773, PMID:27462815
- The eIF-3 complex specifically targets and regulates translation of mRNAs involved in cell proliferation, differentiation, and apoptosis through RNA stem-loop binding, acting as either activator or repressor. Related references: PMID:25849773
- Required for nonsense-mediated mRNA decay (NMD); may cooperate with UPF2 to divert mRNAs from translation to the NMD pathway. Related references: PMID:17468741
- May interact with MCM7 and EPAS1 and regulate their proteasome-mediated degradation. Related references: PMID:17310990, PMID:17324924
- Localizes to the cytoplasm, nucleus, and PML bodies.
- Ubiquitously expressed; highest levels in appendix, lymph, pancreas, skeletal muscle, spleen, and thymus.
- Subject to acetylation and phosphorylation, as determined by proteomics.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 210–350) lies within a conserved region of eIF3e; considering this, the antibody may show cross-reactivity with mouse and rat, but validation in the relevant sample system is recommended.
- For Western blot, the predicted molecular weight is 52 kDa; consider using a standard dilution series and appropriate loading controls.
- IHC-P may require antigen retrieval optimization; run parallel controls to validate specificity.
- ELISA protocols should incorporate antibody pair screening and specificity assays.
