Biological Background: EIF2S3 Function and Localization

  • EIF2S3, also known as eIF2γ, is the gamma subunit of eukaryotic translation initiation factor 2 (eIF2), playing a central role in protein synthesis.
  • As part of the eIF2 complex, it forms a ternary complex with GTP and initiator tRNA (Met-tRNAi), which binds to the 40S ribosomal subunit to initiate translation (PubMed:31836389). Related references: PMID:31836389
  • After mRNA binding and formation of the 43S pre-initiation complex, GTP hydrolysis on eIF2 triggers release of the eIF2-GDP binary complex, allowing 60S subunit joining and 80S initiation complex formation.
  • The GDP-bound eIF2 is recycled to GTP-bound form by the guanine nucleotide exchange factor eIF2B, enabling another round of initiation.
