Biological Background: EIF1AY Function and Localization
- EIF1AY is the Y-chromosomal paralog of EIF1A, also designated eIF-4C, and belongs to the eukaryotic translation initiation factor 1A family.
- It is an essential component of the 43S pre-initiation complex (43S PIC) that binds to the mRNA cap-proximal region and scans the 5' untranslated region to locate the start codon.
- Together with EIF1, EIF1AY facilitates ribosomal scanning, start codon recognition, and assembly of the 48S initiation complex at the initiation codon.
- After start codon recognition, it cooperates with EIF5B to promote joining of the 60S ribosomal subunit and formation of the 80S initiation complex; EIF1AY is released after GTP hydrolysis by EIF5B.
- Its globular domain occupies the ribosomal A site and interacts with EIF5 during scanning to help maintain EIF1 within the open 43S PIC.
- Primarily localized to the cytoplasm, consistent with its role in translation.
- Expressed ubiquitously across human tissues.
