Biological Background: eIF1A (EIF1AX) Function and Localization
- eIF1A, also known as eIF-4C, is a translation initiation factor essential for cap-dependent protein synthesis.
- It enhances formation of the cap-proximal complex, facilitates mRNA scanning, and promotes start codon recognition (PubMed:9732867). Related references: PMID:9732867
- After start codon location, it cooperates with EIF5B to orient initiator Met-tRNA and enable 60S subunit joining; its globular domain occupies the A site of the 40S subunit (PubMed:35732735). Related references: PMID:35732735
- During scanning, eIF1A interacts with EIF5 to help maintain EIF1 in the open 43S PIC, though, unlike its yeast ortholog, it does not bind EIF1 directly (PubMed:24319994). Related references: PMID:24319994
- It localizes to the cytoplasm and participates in host-virus interactions, with RNA-binding and tRNA-binding activities.
- As a reference proteome member, it is widely used in studies of translation control and protein biosynthesis.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 65–144 of human eIF1A, targeting the central region; consider this when designing detection antibodies or epitope mapping.
- Predicted molecular weight is ~16 kDa; for WB, use appropriate gel acrylamide percentage and optimized transfer conditions for low-molecular-weight proteins.
- Cross-reactivity with mouse suggests utility in rodent model studies; always validate performance in your specific sample matrix and experimental system.
- In ELISA applications, consider testing direct vs. sandwich formats and optimize coating antigen concentration and antibody dilutions to maximize signal-to-noise ratio.
