Biological Background: EHHADH Function and Localization
- Encoded by the EHHADH gene, the peroxisomal bifunctional enzyme (also known as L-bifunctional protein, MFE1) is a trifunctional enzyme essential for peroxisomal β-oxidation of long-chain and branched-chain fatty acids.
- Possesses 2-enoyl-CoA hydratase, 3-hydroxyacyl-CoA dehydrogenase, and delta3-delta2-enoyl-CoA isomerase activities, catalyzing two of the four reactions in the pathway (By similarity).
- Together with HSD17B4, catalyzes the hydration of trans-2-enoyl-CoA and dehydrogenation of 3-hydroxyacyl-CoA with opposite chiral specificity, contributing to dicarboxylic acid degradation (PubMed:15060085).

- For immunofluorescence, permeabilization is required to visualize peroxisomal localization; co-staining with peroxisomal markers can confirm specificity.
