Biological Background: EHF Function and Localization
- EHF (ETS homologous factor), also known as ESE-3, is an ETS domain-containing transcription factor with roles in epithelial cell regulation.
- Acts as both transcriptional activator and repressor, binding to consensus GGAA sequences to regulate gene expression and modulate MAP kinase signaling.
- Involved in regulation of TNFRSF10B/DR5 expression through its promoter, implicating functions in apoptosis and tumor suppression.
- May contribute to development and carcinogenesis by acting as a tumor suppressor gene or anti-oncogene.
- Expressed exclusively in epithelial tissues: highly expressed in salivary gland, mammary gland, prostate, and lung; weakly in kidney and colon; absent in most non-epithelial tissues.
- Localizes to the nucleus to exert its DNA-binding and transcriptional regulatory functions.
- Alternative splicing produces multiple isoforms, and the protein is subject to post-translational modifications.
- Keywords: Alternative splicing, DNA-binding, Nucleus, Proteomics identification, Reference proteome, Transcription, Transcription regulation.
Experimental Guidance and Technical Tips
- This antibody was raised against the N-terminal region (aa 1-113) of human EHF. It is recommended for Western blotting and ELISA applications, and may show cross-reactivity with mouse EHF due to sequence conservation.
- For Western blotting, consider using nuclear extracts from epithelial cell lines such as MCF-7 (breast), LNCaP (prostate), or A549 (lung). Validate band specificity using appropriate negative controls and lysates from non-epithelial tissues.
- Optimal antibody dilutions and incubation conditions should be determined empirically for each assay; include known positive and negative samples to confirm reactivity.
