Biological Background: EHD1 Function and Localization
- EH domain-containing protein 1 (EHD1), also known as PAST homolog 1 (hPAST1) or Testilin, is an ATP- and membrane-binding protein that controls membrane reorganization and tubulation upon ATP hydrolysis. Related references: PMID:24019528
- In vitro, EHD1 causes vesiculation of endocytic membranes and acts in early endocytic membrane fusion and trafficking of recycling endosomes. Related references: PMID:15020713 PMID:17233914 PMID:20801876
- Recruited to endosomal membranes upon nerve growth factor stimulation, EHD1 indirectly regulates neurite outgrowth. It also plays a role in myoblast fusion.
- EHD1 is involved in the unidirectional retrograde dendritic transport of endocytosed BACE1 and efficient sorting of BACE1 to axons, implicating a function in neuronal APP processing.
- Plays a role in the formation of the ciliary vesicle (CV), an early step in cilium biogenesis, by recruiting SNARE complex component SNAP29 and is required for recruitment of transition zone proteins. Related references: PMID:31615969 PMID:25686250
- Subcellular localization: recycling endosome membrane, early endosome membrane, cell membrane, and cilium membrane.
- Highly expressed in testis.
- Keywords: ATP-binding, calcium, metal-binding, nucleotide-binding, phosphoprotein, protein transport, acetylation, coiled coil.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1-100 of human EHD1, which may provide high specificity; validate in your sample system.
- For Western blotting, the predicted molecular weight is ~61 kDa; consider using positive controls such as testis lysate due to high expression.
- For immunohistochemistry (paraffin), optimize antigen retrieval conditions as the target is membrane-associated.
- For ELISA, the monoclonal nature may offer consistent performance; verify cross-reactivity with mouse and rat samples as indicated.
- The antibody is recommended for WB, IHC-P, and ELISA; suitability for other applications should be tested empirically.
