Biological Background: EGR1 Function and Localization
- EGR1 functions as a transcriptional regulator, binding to the DNA sequence 5'-GCG(T/G)GGGCG-3' (EGR-site) in target gene promoters. Related references: PMID:20121949
- It binds double-stranded target DNA irrespective of cytosine methylation status, suggesting epigenetic regulatory roles. Related references: PMID:25258363 PMID:25999311
- EGR1 controls expression of genes governing cell survival, proliferation, and apoptosis; it activates tumor suppressors p53 and TGFB1.
- It is required for normal mitosis and hepatocyte proliferation after liver injury.
- EGR1 mediates cellular responses to ischemia/hypoxia by inducing inflammatory cytokines such as IL1B and CXCL2.
- It contributes to circadian clock regulation by modulating the rhythmic expression of core clock genes in the liver and suprachiasmatic nucleus.
- EGR1 protein is abundant in neutrophilic granulocytes.
- Localizes to both nucleus and cytoplasm.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–250) of human EGR1; cross-reactivity with mouse and rat suggests conserved epitopes. Validate species-specific detection in relevant samples.
- For western blot (WB), a band at approximately 58 kDa is expected; use positive controls such as neutrophil or liver lysates.
- For immunofluorescence/ICC, optimize fixation and permeabilization conditions, as EGR1 localizes to both nucleus and cytoplasm.
- For ELISA, consider coating with recombinant EGR1 protein and pairing with a suitable detection antibody; titrate antibody concentrations for optimal signal-to-noise ratio.
- Validate the antibody performance in the intended application and sample type before quantitative experiments.
