Biological Background: EGFR Function and Localization
- Epidermal growth factor receptor (EGFR), also designated Proto-oncogene c-ErbB-1 and Receptor tyrosine-protein kinase erbB-1, is encoded by the EGFR gene (alternative names: ERBB, ERBB1, HER1).
- Functions as a receptor tyrosine kinase binding EGF family ligands (EGF, TGFA, AREG, EPGN, BTC, EREG, HBEGF) to induce receptor dimerization and autophosphorylation, thereby activating major downstream pathways including RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, STATs, and potentially NF-κB (PubMed:10805725, PubMed:27153536, PubMed:2790960, PubMed:12297049, PubMed:15611079, PubMed:17909029, PubMed:11116146). Related references: PMID:10805725 PMID:27153536 PMID:2790960 PMID:12297049 PMID:15611079 PMID:17909029 PMID:11116146
- Directly phosphorylates downstream effectors such as RGS16 (enhancing its GTPase activity and linking to GPCR signaling) and MUC1 (promoting SRC and CTNNB1 interactions), and positively regulates cell migration via interaction with CCDC88A/GIV (PubMed:11602604, PubMed:11483589, PubMed:20462955). Related references: PMID:11602604 PMID:11483589 PMID:20462955
- An alternative splicing isoform (isoform 2) may function as an antagonist of EGF-mediated signaling.
- Subcellular localization includes the cell membrane, endoplasmic reticulum, Golgi apparatus, nucleus, endosomes, and secreted forms, reflecting its dynamic trafficking and signaling roles.
- Ubiquitously expressed in human tissues, with isoform 2 notably detected in ovarian cancers.
- Classified as a proto-oncogene, tyrosine-protein kinase, and transmembrane glycoprotein; also serves as a host cell receptor for hepatitis C virus (HCV) entry, facilitating viral internalization.
- Post-translational modifications include phosphorylation, glycosylation, hydroxylation, methylation, palmitoylation, and ubiquitin-like conjugation, as indicated by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen (aa 1021–1210) lies within the intracellular C-terminal domain; thus the antibody may detect both full-length and C-terminal fragments. Verify specificity in relevant cell lysates or tissue samples.
- For Western blotting, a band around 134 kDa is expected; however, glycosylation and phosphorylation can cause mobility shifts. Include molecular weight markers and validate with EGFR-positive and -negative controls.
- For IHC-P, standard antigen retrieval (e.g., citrate buffer pH 6.0) and blocking protocols should be optimized; test with known positive (e.g., skin, placenta) and negative (e.g., knockout) tissues.
- In IF/ICC, fixation methods (PFA vs. methanol) affect EGFR membrane staining; parallel detection with a membrane marker is recommended.
- For ELISA, the antibody can be used as a capture or detection reagent; assay conditions (coating concentration, blocking) require individual optimization.
- Cross-reactivity is reported for human samples; if used in other species, verify reactivity independently due to potential sequence variations.
