Biological Background: EGFR Function and Localization

  • EGFR is a proto-oncogene member of the ErbB family of receptor tyrosine kinases, also known as HER1 or ErbB-1.
  • Binds a variety of EGF-like ligands (e.g., EGF, TGF-alpha, HB-EGF), leading to receptor dimerization, autophosphorylation, and activation of major signaling cascades including RAS-RAF-MEK-ERK, PI3K-AKT, PLCgamma-PKC, and STATs.   • For Western blot, use denaturing and reducing conditions; expect a band near 134 kDa. Validate using cell lysates expressing EGFR L858R and wild-type controls.
  • For IHC-P, optimize antigen retrieval and include tissue known to harbor the L858R mutation (e.g., lung adenocarcinoma) as a positive control.
  • For ELISA, consider using the immunizing peptide or recombinant EGFR L858R protein as a standard, and assess potential cross-reactivity with wild-type EGFR.
