Biological Background: EGF Function and Localization
- EGF is synthesized as a membrane-anchored precursor protein (Pro-epidermal growth factor) that is proteolytically processed to release mature EGF, which contains multiple EGF-like domains and functions as a potent growth factor.
- EGF stimulates the proliferation and differentiation of epidermal and epithelial tissues both in vivo and in vitro, and also promotes the growth of certain fibroblasts in cell culture.
- Acts as a magnesiotropic hormone in the kidney, enhancing magnesium reabsorption in the distal convoluted tubule through EGFR-mediated activation of the TRPM6 magnesium channel.

- Localized primarily to the cell membrane as a transmembrane protein, with its ectodomain shed to generate soluble mature EGF.

- The immunogen is a synthetic peptide spanning amino acids 851–950 of human EGF (accession NP_001954.2), residing in the C-terminal region of the proprotein. This region may be retained in the membrane-bound precursor and some processed forms.
- For Western blotting (WB), expect major bands corresponding to the full-length precursor (approximately 134 kDa) and possibly lower molecular weight species representing cleavage products or mature EGF. Validate in EGF-expressing samples such as kidney, salivary gland, or prostate tissues.
- In immunofluorescence (IF-P), staining should localize to the plasma membrane, consistent with the membrane-bound precursor. Permeabilization and fixation conditions should be optimized for the specific sample system.
- For ELISA applications, consider pairing this polyclonal antibody with validated capture or detection reagents. Test serial dilutions to establish optimal working concentrations for your specific assay format.
- Cross-reactivity with mouse and rat EGF has been reported; however, antibody performance in these species should be verified individually under your experimental conditions.
