Biological Background: EFTUD2 Function and Localization

  • EFTUD2 (116 kDa U5 snRNP component, SNU114, also known as elongation factor Tu GTP-binding domain-containing protein 2) is a GTP-binding protein that serves as a structural and functional subunit of the U5 small nuclear ribonucleoprotein (snRNP), essential for pre-mRNA splicing. Related references: PMID:16723661
  • It is integral to the U4/U6-U5 tri-snRNP complex, a building block of the major spliceosome, and is required for pre-catalytic, catalytic, and post-catalytic spliceosome assembly and disassembly. Related references: PMID:25092792, PMID:28076346, PMID:28502770
  • EFTUD2 also functions in the minor spliceosome and is probably involved in the splicing of U12-type introns in pre-mRNAs.
  • The protein localizes exclusively to the nucleus, consistent with its primary role in mRNA processing.
  • Post-translational modifications include acetylation and phosphorylation, and it is subject to alternative splicing and ubiquitin conjugation.
  • Mutations in EFTUD2 are linked to mandibulofacial dysostosis with microcephaly (MFDGA), highlighting its developmental importance.

Experimental Guidance and Technical Tips

  • For Western blot analysis, the expected molecular weight of EFTUD2 is approximately 109 kDa (theoretical) or 116 kDa (observed), so ensure proper gel percentage and transfer conditions.
  • In immunofluorescence and immunohistochemistry, anticipate nuclear staining; include DAPI co-staining to confirm localization.
  • For immunoprecipitation, this polyclonal antibody can efficiently pull down native EFTUD2 from human, mouse, and rat cell lysates; consider cross-linking antibody to beads for cleaner results.
  • Because this is a polyclonal antibody, we recommend validating each new lot in your specific assay system and including appropriate positive and negative controls.
  • The immunogen (N-terminal 1–205 amino acids) may not recognize isoforms lacking this region; verify target expression in your model.
  • Optimize antibody dilutions and blocking conditions for each application, as performance can vary between techniques and sample types.
