Biological Background: EFNA1 Function and Localization
- EFNA1 encodes ephrin-A1, a glycosylphosphatidylinositol (GPI)-anchored cell surface ligand of the Eph receptor tyrosine kinase family. It is also known as LERK-1, B61, and TNF alpha-induced protein 4.
- Ephrin-A1 binds promiscuously to Eph receptors on adjacent cells, mediating contact-dependent bidirectional signaling that regulates cell migration, repulsion, and adhesion during neuronal, vascular, and epithelial development. Related references: PMID:17332925 PMID:18794797
- In vascular biology, ephrin-A1 plays a critical role in angiogenesis by activating EPHA2 to promote endothelial cell differentiation and migration.
- As a ligand for EPHA3, ephrin-A1 inhibits epithelial-to-mesenchymal transition in cardiac cells, contributing to proper heart development. Related references: PMID:12794130
- Ephrin-A1 may also participate in dendritic spine morphogenesis (by similarity).
- The protein localizes predominantly to the cell membrane via a GPI anchor but can be released as a soluble form through cleavage. It is abundantly expressed in brain, and its expression is often down-regulated in primary glioma. Soluble monomeric ephrin-A1 has been detected in glioblastoma and breast cancer cells.
- Post-translational modifications include disulfide bond formation and N-linked glycosylation. The protein undergoes alternative splicing, generating membrane-bound and secreted isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 19-182 of human EFNA1, covering the extracellular receptor-binding domain. The resulting polyclonal antibody may recognize both full-length GPI-anchored and soluble isoforms.
- For Western blot, users may test antibody dilutions in the range commonly used for polyclonal antibodies (e.g., 1:500–1:2000) and confirm specificity using lysates from EFNA1-overexpressing and knockdown cell lines. Endogenous EFNA1 migrates at approximately 24 kDa.
- For IHC-P, consider optimizing antigen retrieval and include appropriate tissue controls. Both paraffin-embedded human and mouse tissue sections can be used, given the claimed cross-reactivity.
- In ELISA, the antibody can be employed as a capture or detection reagent; the optimal working concentration should be determined experimentally in the specific assay format.
- Due to the GPI-anchored nature and potential shedding of ephrin-A1, detectability may vary between cell types and treatment conditions; validation in the relevant sample system is advised.
