Biological Background: eEF1A1 Function and Localization
- eEF1A1 (Elongation factor 1-alpha 1) is a highly conserved translation elongation factor, also known as EF1A, EF-Tu, and GRAF-1EF.
- It catalyzes the GTP-dependent binding of aminoacyl-tRNA to the ribosomal A-site during protein synthesis elongation. Related references: PMID:26593721, PMID:26651998, PMID:36123449
- eEF1A1 also functions as a positive regulator of IFNG transcription in T-helper 1 cells by participating in a promoter-binding complex with TXK and PARP1. Related references: PMID:17177976
- Beyond translation, it plays a role in cytoskeleton organization by promoting actin bundling (By similarity).
- The protein is predominantly cytoplasmic but is also found in the nucleus, nucleolus, and associated with the cell membrane.
- eEF1A1 is subject to various post-translational modifications including acetylation, methylation, and phosphorylation, and is required for the translation of viral proteins during SARS-CoV-2 infection.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from aa 301–400 of human eEF1A1, targeting a conserved region with expected cross-reactivity to mouse and rat.
- For Western blot (WB), the predicted molecular weight is ~50 kDa; verify band specificity using appropriate positive and negative controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval conditions and use species-matched detection systems.
- In ELISA, titrate the antibody and validate against the immunogen peptide or recombinant protein standard.
- Cross-reactivity with human, mouse, and rat makes it suitable for multi-species studies; however, performance in other species should be validated.
