Biological Background: eEF1A1 Function and Localization
- eEF1A1 (Elongation factor 1-alpha 1) is a ubiquitously expressed protein that belongs to the GTP-binding elongation factor family. It is also known as EF-Tu, eukaryotic elongation factor 1 A-1 (eEF1A-1), and leukocyte receptor cluster member 7.
- As a core component of the translational machinery, eEF1A1 catalyzes the GTP-dependent binding of aminoacyl-tRNA to the A-site of ribosomes during protein synthesis elongation, facilitating codon-anticodon recognition and peptide bond formation. - eEF1A1 also plays a non-canonical role in transcriptional regulation, positively modulating IFNG expression in T-helper 1 cells by forming a complex with TXK and PARP1 at the IFNG promoter. Related references: PMID:17177976
- The protein is involved in cytoskeleton organization by promoting actin bundling.
- Subcellularly, eEF1A1 localizes primarily to the cytoplasm, but is also found in the nucleus, nucleolus, and at the cell membrane.
- During SARS-CoV-2 infection, eEF1A1 is required for the translation of viral proteins and efficient viral replication.
- Post-translational modifications of eEF1A1 include acetylation, methylation, phosphorylation, and ubiquitination, as identified through proteomics approaches.
