Biological Background: eEF1A1 Function and Localization
- The gene is a member of the GTP-binding elongation factor family and is also known as EF-Tu, eEF1A-1, and LENG7.
- eEF1A1 catalyzes the GTP-dependent binding of aminoacyl-tRNA (aa-tRNA) to the A-site of ribosomes during the elongation phase of protein synthesis. Related references: PMID:26593721 PMID:26651998 PMID:36123449
- In addition to its canonical role in translation, eEF1A1 positively regulates IFNG transcription in T-helper 1 cells as part of a promoter-binding complex with TXK and PARP1. Related references: PMID:17177976
- The protein also contributes to cytoskeleton organization by promoting actin bundling.
- eEF1A1 is primarily localized in the cytoplasm but can also be found in the nucleus, nucleolus, and associated with the cell membrane.
- Post-translational modifications include acetylation, methylation, phosphorylation, and isopeptide bond formation, reflecting its multifaceted regulation.
- During SARS-CoV-2 infection, eEF1A1 is required for the translation of viral proteins and viral replication.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 123–462 of human eEF1A1; the antibody is expected to detect the full-length protein at ~50 kDa in WB, but validation in the relevant sample system is recommended.
- For IF/ICC, since eEF1A1 localizes to both cytoplasm and nucleus, permeabilization may be necessary; consider optimizing fixation and blocking conditions.
- As a polyclonal antibody, background may be observed in some applications; titration and appropriate controls are advised.
- Verified cross-reactivity with mouse and rat supports use in common model organisms, but independent confirmation for each species is prudent.
