Biological Background: Endothelin-1 Function and Localization
- Endothelin-1 is encoded by the EDN1 gene and produced as a preproprotein (PPET1) that undergoes proteolytic processing to yield the mature 21-amino acid vasoactive peptide.
- It belongs to the endothelin family, which are potent vasoconstrictors primarily secreted by endothelial cells.
- Endothelin-1 acts as a ligand for G protein-coupled receptors EDNRA and EDNRB, leading to activation of PTK2B, BCAR1, BCAR3, and GTPases RAP1 and RHOA in glomerular mesangial cells. Related references: PMID:19086031

- By similarity, endothelin-1 promotes mesenteric arterial wall remodeling through ROCK signaling, leading to NFATC3 nuclear translocation and upregulation of ACTA2, a marker of smooth muscle hypertrophy.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 18-212 of human EDN1, covering the preproprotein region. The antibody may recognize both precursor and mature forms; consider this when analyzing bands in Western blot.
- Predicted molecular weight of the preproprotein is 24 kDa; mature endothelin-1 is approximately 2.5 kDa. Optimize gel percentage and transfer conditions for small peptides if needed.
- For IF/ICC, validate permeabilization and fixation conditions, as endothelin-1 is secreted; consider co-staining with organelle markers.
- When using for ELISA, pair with an appropriate detection antibody and validate in the relevant sample matrix (e.g., serum, plasma) to avoid interference.
