Biological Background: Endothelin-1 Function and Localization

  • Endothelin-1 is encoded by the EDN1 gene and produced as a preproprotein (PPET1) that undergoes proteolytic processing to yield the mature 21-amino acid vasoactive peptide.
  • It belongs to the endothelin family, which are potent vasoconstrictors primarily secreted by endothelial cells.
  • Endothelin-1 acts as a ligand for G protein-coupled receptors EDNRA and EDNRB, leading to activation of PTK2B, BCAR1, BCAR3, and GTPases RAP1 and RHOA in glomerular mesangial cells. Related references: PMID:19086031
