Experimental Guidance and Technical Tips
- Given the immunogen design (N-terminal peptide, aa 1–100), the antibody is expected to recognize an epitope in the extracellular N-terminus of LPAR1; verify specificity using relevant controls.
- For Western blot, LPAR1 is a multi-pass membrane protein with a predicted molecular weight of ~41 kDa; inclusion of mild detergents in lysis buffer may be necessary to solubilize the receptor.
- In ELISA applications, consider using the immunogen peptide as a positive control to assess antibody binding.
- Due to the GPCR nature, post-translational modifications such as glycosylation may cause band shifts; deglycosylation treatment can be used to confirm target identity.
- Validate antibody performance in the appropriate species (human) and cell line expressing LPAR1; cross-reactivity with other species has not been determined.
