Biological Background: EDF1 Function and Localization

  • EDF1 (Endothelial differentiation-related factor 1), also known as Multiprotein-bridging factor 1 (MBF1), is a 16 kDa transcriptional coactivator encoded by the EDF1 gene on chromosome 9.
  • Functions as a transcriptional coactivator, enhancing the transcriptional activities of NR5A1 and ligand-dependent NR1H3/LXRA and PPARG, and increasing DNA-binding activity of ATF1, ATF2, CREB1, and NR5A1.
  • Regulates nitric oxide synthase activity likely by sequestering calmodulin in the cytoplasm.
  • Involved in endothelial cell differentiation, hormone-induced cardiomyocyte hypertrophy, and lipid metabolism.
  • Localizes to both the cytoplasm and nucleus.
  • Ubiquitously expressed, with higher abundance in heart, pancreas, liver, intestine, and adipose tissues.
  • Subject to post-translational modifications including acetylation, methylation, and phosphorylation; also undergoes alternative splicing.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein covering amino acids 1-148 of human EDF1 (NP_003783.1). For optimal results, consider validating the antibody in your specific sample system.
  • This antibody has been tested in Western blot (WB), immunohistochemistry (IHC-P), and ELISA. Cross-reactivity is expected with human, mouse, and rat due to sequence conservation.
  • When performing WB, the predicted molecular weight of EDF1 is ~16 kDa. Verify band size with appropriate molecular weight markers.
  • For IHC-P, antigen retrieval conditions may need optimization based on tissue type and fixation method.
