Biological Background: EDEM2 Function and Localization
- EDEM2 (C20orf31, C20orf49, bA4204.1) is an ER degradation-enhancing alpha-mannosidase-like protein 2.
- It plays a key role in the endoplasmic reticulum-associated degradation (ERAD) pathway, recognizing and targeting misfolded glycoproteins for proteasomal degradation in an N-glycan-dependent manner. Related references: PMID:15537790 PMID:25092655
- EDEM2 is proposed to initiate ERAD by catalyzing the first mannose trimming step of substrate glycans, converting Man9GlcNAc2 to Man8GlcNAc2. Related references: PMID:25092655
- It likely binds to exposed hydrophobic patches on misfolded proteins, facilitating their retrotranslocation and degradation.
- The protein localizes to the lumen of the endoplasmic reticulum.
- EDEM2 is ubiquitously expressed across human tissues, with strongest expression in small intestine and peripheral blood leukocytes and lowest in brain and skeletal muscle.
- It is subject to alternative splicing and participates in the unfolded protein response.
- The predicted molecular weight of human EDEM2 is approximately 65 kDa.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant fragment covering amino acids 399–578 of human EDEM2, a region that may confer target specificity; consider verifying with your own controls.
- For Western blot, a band around 65 kDa is expected; using cell lines such as HeLa or HEK293 as positive controls is advisable, but validation in your specific sample system is recommended.
- In immunocytochemistry (IF/ICC), fixation and permeabilization conditions should be optimized for ER luminal proteins; initial testing with known EDEM2-expressing cells can help establish the protocol.
- ELISA applications can be developed using recombinant EDEM2 protein or cell-based assays; titrate the antibody to determine the optimal working concentration.
- The antibody cross-reacts with mouse EDEM2, supporting its use in mouse model studies; however, always confirm reactivity with appropriate mouse tissue lysates.
- As a polyclonal antibody, lot-to-lot variability may occur; include positive and negative controls in each experiment.
