Biological Background: ECI1 Function and Localization
- ECI1 (Enoyl-CoA delta isomerase 1, mitochondrial), also known as DCI or dodecenoyl-CoA isomerase, is a mitochondrial enzyme encoded by the ECI1 gene.
- It functions as an auxiliary enzyme in the beta-oxidation of mono- and polyunsaturated fatty acids, catalyzing the isomerization of 3-cis and 3-trans double bonds to the 2-trans form to allow re-entry into the beta-oxidation cycle. Related references: PMID:26474213, PMID:7818490
- Together with 2,4-dienoyl-CoA reductase (DECR) and Delta(3,5)-Delta(2,4)-dienoyl-CoA isomerase (ECH1), ECI1 ensures efficient degradation of unsaturated fatty acids. Related references: PMID:26474213
- Subcellularly localized to the mitochondrial matrix, consistent with its role in fatty acid metabolism.

- For WB, load appropriate positive control lysates (e.g., human liver) and validate the ~33 kDa band; include a mitochondrial fraction if needed.
- For IHC-P and IF/ICC, optimize antigen retrieval and use human liver sections as a positive control given the high hepatic expression.
- The antibody cross-reacts with mouse, allowing use in rodent models, but validate in the relevant sample system as performance may vary.
