Biological Background: EBP1/PA2G4 Function and Localization
- EBP1, encoded by the PA2G4 gene, is also known as proliferation-associated protein 2G4, ErbB3-binding protein 1, and cell cycle protein p38-2G4 homolog (hG4-1). It exists as at least two isoforms generated by alternative splicing; isoform 1 suppresses apoptosis, while isoform 2 promotes cell differentiation.
- The protein plays a role in ERBB3-regulated signal transduction pathways, acting as a corepressor of the androgen receptor (AR) and inhibiting transcription of certain E2F1-regulated promoters, likely through recruitment of histone acetylase (HAT) activity.
- EBP1 binds RNA and associates with mature rRNAs (28S, 18S, 5.8S) and rRNA precursors, suggesting involvement in ribosomal RNA processing and ribosome assembly. It may also mediate cap-independent translation of viral IRES elements.
- Subcellular localization includes cytoplasm, nucleus, and nucleolus, consistent with its roles in signal transduction, transcriptional regulation, and ribosome biogenesis.
- The protein is regulated by phosphorylation, acetylation, and ubiquitination, as indicated by keywords "Phosphoprotein", "Acetylation", and "Ubl conjugation". A 3D structure is available in the PDB.
- Isoform 1 is strongly expressed in cancer cells, while isoform 2 is undetectable at the protein level in these cells. Both isoforms are widely expressed in normal tissues, including heart, brain, lung, pancreas, skeletal muscle, kidney, placenta, and liver.
