Biological Background: EBP1/PA2G4 Function and Localization
- Also known as Proliferation-associated protein 2G4, ErbB3-binding protein 1 (EBP1), or cell cycle protein p38-2G4 homolog; encoded by the PA2G4 gene; exists as two major isoforms (isoform 1 and isoform 2) via alternative splicing.
- Functions in ERBB3-regulated signal transduction, acting as a corepressor of the androgen receptor (AR) and inhibiting E2F1-dependent transcription, likely by recruiting histone acetylase (HAT) activity.
- Binds RNA and associates with mature rRNAs (28S, 18S, 5.8S), rRNA precursors, and U3 snoRNA; implicated in intermediate and late steps of rRNA processing and ribosome assembly.
- Mediates cap-independent translation of specific viral mRNAs carrying internal ribosomal entry sites (IRESs), contributing to host translation regulation.
- Regulates cell proliferation, differentiation, and survival; isoform 1 suppresses apoptosis while isoform 2 promotes cell differentiation.
- Localizes to the cytoplasm, nucleus, and nucleolus, consistent with its roles in signaling, transcription, and ribosome biogenesis.
- Widely expressed in tissues including heart, brain, lung, pancreas, skeletal muscle, kidney, placenta, and liver; isoform 1 is strongly expressed in cancer cells, whereas isoform 2 is typically undetectable in cancer.
- Subject to post-translational modifications including acetylation, phosphorylation, and ubiquitination, which may modulate its activity and stability.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 300–394 of human PA2G4, a region conserved in mouse and rat, consistent with the antibody’s cross-reactivity.
- For Western blot (WB), a band of approximately 44 kDa is expected; validate detection using a cell line expressing the target (e.g., HeLa, A549).
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider antigen retrieval conditions and inclusion of isotype controls; nuclear/nucleolar staining may be observed.
- For ELISA and other quantitative applications, optimize antibody dilution and confirm linearity using recombinant PA2G4 protein.
