Biological Background: EB3/MAPRE3 Function and Localization
- Microtubule-associated protein RP/EB family member 3 (EB3/MAPRE3), also known as EB1 protein family member 3 (EBF3) or End-binding protein 3 (EB3), is a plus-end tracking protein (+TIP) that binds microtubule plus-ends and promotes microtubule growth. Related references: PMID:19255245 PMID:28814570
- It stabilizes microtubules and anchors them at centrosomes, contributing to spindle function during mitosis and cell division. Related references: PMID:19255245 PMID:28814570
- As a regulator of minus-end microtubule organization, EB3/MAPRE3 interacts with the AKAP9-PDE4DIP complex to recruit CAMSAP2 to the Golgi apparatus, tethering non-centrosomal microtubules and enabling polarized cell movement. Related references: PMID:28814570
- EB3/MAPRE3 promotes elongation of CAMSAP2-decorated microtubule stretches on the minus-end of microtubules. Related references: PMID:28814570
- Predominantly expressed in brain and muscle tissues, with localization to the cytoplasm and cytoskeleton.
- The protein undergoes alternative splicing and phosphorylation, and belongs to the EB1 family of microtubule-associated proteins.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 1–281 of human EB3/MAPRE3, which represents nearly the full-length protein. This design may support recognition of the endogenous target across multiple applications.
- For Western blotting, a band at approximately 32 kDa is expected based on the predicted molecular weight; validation in the relevant sample system is recommended.
- Cross-reactivity with mouse is reported; consider performing validation experiments if mouse samples are to be used.
- The antibody is also suitable for ELISA; users may consider optimizing coating and detection conditions for their specific assay format.
