Biological Background: EAAT1/SLC1A3 Function and Localization
- Excitatory amino acid transporter 1 (EAAT1), also known as SLC1A3 or GLAST-1, is a member of the solute carrier family 1 (SLC1) of high-affinity glutamate transporters.
- EAAT1 mediates sodium-dependent, high-affinity uptake of L-glutamate, L-aspartate, and D-aspartate, playing a critical role in clearing glutamate from the synaptic cleft to terminate neurotransmission (PubMed:20477940, PubMed:26690923, PubMed:28032905). - The transporter functions as a symporter, co-transporting one amino acid with two or three Na+ ions and one proton, coupled to the counter-transport of one K+ ion (PubMed:20477940). Related references: PMID:20477940
- EAAT1 also mediates uncoupled chloride flux, which may counteract charge accumulation during aspartate and Na+ symport (PubMed:20477940). Related references: PMID:20477940
- The protein is localized to the cell membrane, consistent with its role in extracellular glutamate clearance.
- EAAT1 is predominantly expressed in the cerebellum, frontal cortex, hippocampus, and basal ganglia of the brain, with very low levels in peripheral tissues such as heart, lung, placenta, and skeletal muscle.
- Post-translational modifications include N-linked glycosylation and phosphorylation, as indicated by UniProt keywords "Glycoprotein" and "Phosphoprotein".

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 150-250 of human EAAT1/SLC1A3; this mid-region epitope is predicted to detect total EAAT1 protein.
- For Western blot (WB) validation, use brain lysates (especially cerebellum) as positive controls, where EAAT1 is highly expressed; expect a band around 60 kDa.
- For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), optimize fixation and permeabilization conditions, as EAAT1 is a multi-pass transmembrane protein.
- This antibody shows cross-reactivity with mouse and rat EAAT1 based on supplier data; however, independent validation in the intended species and sample type is recommended.
- The antibody is also suitable for ELISA applications; consider using it in sandwich ELISA formats with a matched detection antibody pair.
