- E2F4 is a member of the E2F family of transcription factors, playing a crucial role in cell cycle regulation and transcriptional control.
- It binds DNA cooperatively with DP proteins at E2 recognition sites (5'-TTTC[CG]CGC-3') found in promoters of cell cycle and DNA replication genes.
- Functions as a transcription activator, driving the G1 to S phase transition and coordinating the expression of genes required for DNA synthesis.
- Binds with high affinity to pocket proteins RBL1 and RBL2, and can also interact with RB1 under certain conditions.
- Specifically required for multiciliate cell differentiation: together with MCIDAS and E2F5, it activates genes essential for centriole biogenesis.
- Localizes to the nucleus, consistent with its role in transcriptional regulation.
- Widely expressed across human tissues, including heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas.
