Biological Background: E2F1 Function and Localization
- E2F1 is a transcription factor that forms heterodimers with DP proteins to bind E2 recognition sites (5'-TTTC[CG]CGC-3') in target promoters. Related references: PMID:10675335, PMID:12717439, PMID:17050006
- The DRTF1/E2F complex regulates the G1/S transition by controlling expression of genes involved in cell cycle progression and DNA replication. Related references: PMID:10675335, PMID:12717439, PMID:17704056
- E2F1 interacts with the retinoblastoma protein RB1 in a cell-cycle-dependent manner, modulating its transcriptional activity. Related references: PMID:10675335, PMID:12717439, PMID:17704056
- Acts as a dual regulator, promoting both cell proliferation and TP53/p53-dependent apoptosis. Related references: PMID:8170954
- Directly activates transcription of PEG10 and RRP1B, and blocks adipocyte differentiation by repressing CEBPA binding to its target gene promoters. Related references: PMID:17050006, PMID:18625225, PMID:28992046, PMID:20040599, PMID:20176812
- Localizes primarily to the nucleus.
- Post-translationally modified by phosphorylation, acetylation, methylation, and ubiquitination, which regulate stability and activity.
Experimental Guidance and Technical Tips
- The predicted molecular weight of E2F1 is 47 kDa; for Western blotting, consider using an appropriate gel percentage and blocking conditions for this size range.
- For IF/ICC, effective nuclear permeabilization is critical; optimize fixation and permeabilization steps to ensure antibody access to the nuclear compartment.
- The immunogen corresponds to amino acids 200–301 of human E2F1; while cross-reactivity with mouse and rat is reported, validate detection specificity in your target species and tissue.
- This polyclonal antibody may recognize multiple E2F1 isoforms; confirm band patterns using appropriate controls and, if necessary, siRNA knockdown.
- For ELISA applications, consider checkerboard titration to determine optimal coating and detection antibody concentrations.
