Biological Background: E2A/TCF3 Function and Localization
- E2A/TCF3 encodes two predominant isoforms, E12 and E47, generated by alternative splicing; both belong to the class B basic helix-loop-helix (bHLH) transcription factor family.
- It heterodimerizes with tissue-specific bHLH proteins to bind E-box motifs (5'-CANNTG-3') and specific enhancer elements, such as the kappa-E2 site in the immunoglobulin gene enhancer, thereby regulating cell fate in embryogenesis and B-cell differentiation (By similarity). - Together with TCF15, it is required for mesenchymal to epithelial transition (MET) (By similarity).
- It facilitates ATOH7 binding to DNA at the consensus sequence 5'-CAGGTG-3' and positively regulates its transcriptional activity.
- Subcellular localization is strictly nuclear, consistent with its role as a transcription factor.
- Post-translational modifications include phosphorylation, methylation, and ubiquitin-like conjugation; it is also classified as a proto-oncogene due to its involvement in chromosomal rearrangements.
