Biological Background: E2A Function and Localization
- E2A (Transcription factor E2-alpha), encoded by the TCF3 gene, is a member of the class B basic helix-loop-helix (bHLH) family (bHLHb21) and is also known as ITF1, E47, and p75.
- Acts as a transcriptional regulator involved in the initiation of neuronal differentiation and facilitates the mesenchymal to epithelial transition during development.
- Heterodimerizes with tissue-specific bHLH proteins to determine cell fate; for example, it plays essential roles in muscle and early B-cell differentiation.
- Binds DNA at E-box motifs (5'-CANNTG-3') and recognizes the kappa-E2 site in the immunoglobulin gene enhancer to regulate B-cell-specific gene expression. Related references: PMID:2493990
- Localizes predominantly to the nucleus, consistent with its function as a transcription factor.
- Undergoes alternative splicing and is subject to post-translational modifications including phosphorylation, methylation, and ubiquitination, which may modulate its activity.
Experimental Guidance and Technical Tips
- The immunogen was designed against amino acids 450–525 of human E2A; consider validating the antibody's reactivity with full-length and splice variant proteins in your specific sample system.
- In western blotting (WB), a band at approximately 68 kDa is anticipated; using whole-cell lysates from Jurkat or HeLa cells may serve as positive controls.
- For immunohistochemistry (IHC-P), antigen retrieval and optimization of antibody dilution are recommended; tissue-specific expression should be verified with known positive and negative tissues.
- Cross-reactivity with mouse and rat is indicated; when using these species, it is advisable to confirm specificity and sensitivity under your experimental conditions.
