Biological Background: E-Selectin/CD62E Function and Localization

  • E-selectin (CD62E, ELAM-1, LECAM2) is a cell surface glycoprotein encoded by the Sele gene in mice. It belongs to the selectin family of adhesion molecules and functions primarily in immune surveillance and inflammation.
  • Core function: Mediates the initial adhesion and rolling of blood neutrophils on cytokine-activated endothelial cells by interacting with its ligand PSGL-1 (SELPLG). Related references: PMID:1375914
  • May also contribute to capillary morphogenesis, suggesting a role in angiogenesis and new blood vessel formation.
  • Predominantly localized to the cell membrane as a type I transmembrane protein; its extracellular region can be shed upon activation.
  • Structural domains include an N-terminal C-type lectin domain, an EGF-like domain, multiple Sushi (SCR/CCP) repeats, a single transmembrane helix, and a short cytoplasmic tail. Calcium binding to the lectin domain is essential for ligand recognition.
  • Contains multiple disulfide bonds that stabilize the extracellular domains and is extensively glycosylated.
  • Along with L-selectin and P-selectin, it forms the selectin family, each exhibiting distinct but overlapping patterns of expression and ligand specificity.

Experimental Guidance and Technical Tips

  • This rabbit polyclonal antibody was raised against a recombinant protein spanning the extracellular domain (aa 22-557) of mouse E-selectin; it is expected to recognize both the full-length membrane-bound form and the soluble shed ectodomain.
  • For Western blot, a band at approximately 66 kDa (predicted molecular weight ~66.7 kDa) should be observed; be aware that glycosylation may cause slight migration shifts. Titration of both primary and secondary antibodies is recommended.
  • In ELISA applications, the antibody can be used for capture or detection of recombinant or endogenous E-selectin. Validate with appropriate positive and negative controls in your assay system.
  • The product is validated for reactivity with mouse samples; cross-reactivity with other species has not been determined.
  • Because the immunogen covers most of the extracellular region, it may be suitable for detecting denatured protein under reducing conditions; however, native conformation-dependent applications may require empirical optimization.
  • Consider using a 5% non-fat milk or BSA blocking solution to reduce non-specific binding, especially for Western blot of tissue lysates.
