Biological Background: E-Cadherin Function and Localization

  • E-Cadherin (CDH1) is a calcium-dependent cell adhesion protein that mediates homophilic interactions essential for tissue integrity and cell sorting. It also plays a role in regulating epithelial cell mobility and proliferation. Related references: PMID:11976333

  • It is a key component of adherens junctions and contributes to the early formation of desmosomes by facilitating recruitment of DSG2 and DSP. Related references: PMID:29999492

  • The protein possesses a potent invasive suppressor function and acts as a ligand for integrin alpha-E/beta-7.

  • E-Cadherin also promotes non-amyloidogenic degradation of Abeta precursors and inhibits APP C99 and C83 production.

  • It serves as a receptor for Listeria monocytogenes internalin A, mediating bacterial uptake.

  • Subcellularly, it localizes to the cell membrane, adherens junctions, desmosomes, and can also be found in the Golgi, endosomes, and as a secreted fragment following proteolytic cleavage.

  • Tissue expression is prominent in the epithelial layers of skin, esophagus, and liver, as well as in granuloma macrophages.

  • Post-translational modifications include phosphorylation, glycosylation, disulfide bond formation, and ubiquitination, which regulate its stability and function.

Experimental Guidance and Technical Tips

  • The immunogen (aa 373–622) lies within the extracellular domain of E-Cadherin, making this antibody suitable for detecting the full-length transmembrane protein in western blot (WB) at ~97 kDa.
  • For immunohistochemistry on paraffin-embedded sections (IHC-P), antigen retrieval may be necessary; validate with known positive and negative control tissues.
  • ELISA performance can be optimized using recombinant E-Cadherin protein; consider pairing with a detection antibody for quantitative assays.
  • Predicted cross-reactivity with mouse and rat E-Cadherin, but always confirm specificity in the relevant experimental system.
