Biological Background: Cadherin-1 Function and Localization

  • Cadherin-1 (also known as E-cadherin, Uvomorulin, ARC-1) is a calcium-dependent cell adhesion protein that mediates homophilic cell-cell interactions, contributing to the sorting of heterogeneous cell types and regulating epithelial cell adhesion, mobility, and proliferation. Related references: PMID:11976333
  • It promotes the organization of radial actin fiber structure and cellular response to contractile forces by anchoring radial actin fibers to junction complexes at the cell membrane via interaction with AMOTL2.
  • CDH1 plays a role in the early stages of desmosome cell-cell junction formation by facilitating the recruitment of DSG2 and DSP to desmosome plaques.
  • It acts as a potent invasion suppressor and serves as a ligand for integrin alpha-E/beta-7. Additionally, the E-Cad/CTF2 fragment promotes non-amyloidogenic degradation of Abeta precursors and inhibits APP C99 and C83 production.
  • In mice, CDH1 is expressed in non-neural epithelial tissues, with specific detection in inner and outer pillar cells of the organ of Corti, granuloma macrophages, and epidermal keratinocytes from birth onward. Related references: PMID:30639848, PMID:27760340, PMID:18079750, PMID:27375112, PMID:30660666
  • It localizes to the cell membrane, adherens junctions, desmosomes, endosomes, and the Golgi apparatus trans-Golgi network, with cytoplasmic localization also observed.
  • Post-translational modifications include signal peptide cleavage, disulfide bond formation, phosphorylation, and ubiquitin conjugation, consistent with its dynamic role in cell adhesion and signaling.
  • Notably, this mouse cadherin does not function as a receptor for Listeria monocytogenes internalin A without a specific surface residue mutation.

Experimental Guidance and Technical Tips

  • The antibody is generated against recombinant mouse E-Cadherin protein; verify specificity in your experimental system using appropriate positive and negative controls.
  • For IHC-P, consider optimizing antigen retrieval and antibody dilution to achieve clear membrane staining in epithelial tissues from human, mouse, or rat samples.
  • In ELISA, use as a capture or detection antibody after confirming linearity and sensitivity with recombinant protein or cell lysates.
  • Given its cross-reactivity with human, mouse, and rat, this antibody is suitable for comparative studies across species; however, validate expression levels in each species separately.
  • Because E-Cadherin undergoes proteolytic processing, be aware of potential band shifts if exploring other applications; the full-length protein is ~98kDa.
