Biological Background: DYNLL1 Function and Localization
- Component of cytoplasmic dynein 1, a motor protein complex that drives retrograde transport of vesicles and organelles along microtubules.
- Acts as a non-catalytic accessory subunit linking dynein to cargos and adapter proteins, thereby regulating dynein motor function.
- Functions as a protein-protein adapter that inhibits and/or sequesters target proteins, including neuronal nitric oxide synthase (NOS1) and BCL2L11. Related references: PMID:10198631 PMID:15193260 PMID:15891768
- Key regulator of the DNA damage response: upon phosphorylation at Ser-88, recruited by TP53BP1 to DNA double-strand breaks (DSBs) to disrupt MRE11 dimerization and inhibit DNA end resection. In a subset of DSBs, unphosphorylated DYNLL1 promotes recruitment of the Shieldin complex. Related references: PMID:30464262 PMID:37696958
- Mediates apoptosis by sequestering BCL2L11 to microtubules; upon apoptotic stimuli, the BCL2L11-DYNLL1 complex dissociates and translocates to mitochondria, where it sequesters BCL2 and neutralizes its anti-apoptotic activity. Related references: PMID:10198631 PMID:15193260
- Enhances estrogen receptor alpha (ESR1) transactivation and helps direct its nuclear localization. Related references: PMID:15891768 PMID:16684779
- Ubiquitously expressed in tissues, with prominent expression detected in testis.
- Subcellular distribution includes the cytoplasm, cytoskeleton (centrosome, microtubules), nucleus, chromosomes, and mitochondria.
- Post-translational modifications include phosphorylation and acetylation, regulating its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-89 of human DYNLL1, targeting an N-terminal region. Consider this when designing epitope mapping or competition assays.
- For western blotting (WB), the predicted molecular weight is approximately 10 kDa. Use high-percentage SDS-PAGE gels and suitable membrane pore size for efficient transfer of low-molecular-weight proteins.
- For immunohistochemistry (IHC-P), antigen retrieval conditions may require optimization depending on tissue type and fixation method.
- ELISA protocols may benefit from cross-reactivity testing against the synthetic peptide immunogen as a positive control.
- Validate antibody specificity in relevant biological systems, such as DYNLL1 knockout or knockdown cells, to confirm target recognition.
