Biological Background: DYNC2H1 Function and Localization

  • Cytoplasmic dynein 2 heavy chain 1 (DYNC2H1), also known as dynein heavy chain 11 (hDHC11) or dynein heavy chain isotype 1B, is a large motor protein of approximately 493 kDa.
  • It is a key component of the dynein-2 complex and functions as a motor for intraflagellar retrograde transport, essential for cilia biogenesis.
  • DYNC2H1 may also play roles in intracellular transport between the endoplasmic reticulum and Golgi, as well as in Golgi organization.
  • The protein is expressed in multiple isoforms due to alternative splicing.
  • Subcellularly, DYNC2H1 localizes to the cytoplasm, cytoskeleton (cilium axoneme), and cell membrane.
  • Conserved domains include ATP-binding and nucleotide-binding regions, along with coiled-coil domains characteristic of dynein heavy chains.
  • Mutations in DYNC2H1 are associated with ciliopathies, including short-rib thoracic dysplasia and related skeletal disorders, highlighting its developmental importance.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-250), which may allow detection of multiple DYNC2H1 isoforms if the epitope is conserved across splice variants.
  • Given the large molecular weight (~493 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6% or gradient gels) and extended transfer times for western blotting. Ensure adequate protein loading and use appropriate molecular weight markers for high-range detection.
  • For ELISA, the antibody can be used for screening applications; optimize coating and detection conditions based on experimental requirements.
  • Cross-reactivity with rat samples is reported; however, performance in other species should be independently validated.
  • Inclusion of appropriate positive and negative controls is recommended to confirm specificity in your experimental system.
