Biological Background: DYNC1I1 Function and Localization
- Encodes cytoplasmic dynein 1 intermediate chain 1, also known as dynein intermediate chain 1 (DH IC-1), a non-catalytic component of the cytoplasmic dynein 1 motor complex.
- Acts as an accessory component linking dynein to cargos and adapter proteins; specifically mediates binding of dynein to dynactin via the p150-glued (DCTN1) subunit.
- Functions in intracellular retrograde transport of vesicles and organelles along microtubules, and may mediate interaction with membranous organelles and kinetochores.
- Localizes to the cytoplasm and is detected at chromosome centromeres, kinetochores, and spindle poles during cell division.
- Belongs to the dynein intermediate chain family and contains WD repeat domains; subject to post-translational modifications including acetylation and phosphorylation.
- Alternative splicing produces multiple isoforms; implicated in transport and motor protein functions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1-220 amino acids of human DYNC1I1; this region may be conserved across human, mouse, and rat, consistent with observed cross-reactivity.
- For Western blotting, the expected band is around 73 kDa; validate in relevant cell lysates and consider using positive and negative controls.
- IHC-P may require antigen retrieval and optimization of antibody dilution; nuclear/centrosomal staining patterns should be confirmed with subcellular markers.
- ELISA performance can be assessed with recombinant protein or peptide antigens; adjust coating and detection conditions empirically.
