Biological Background: DVL1 Function and Localization
- DVL1 is a segment polarity protein dishevelled homolog, also known as DSH homolog 1, encoded by the DVL1 gene. It is a key component of the Wnt signaling pathway.
- Participates in both canonical and non-canonical Wnt signaling by binding to the cytoplasmic C-terminus of frizzled receptors and transducing signals to downstream effectors.
- Mediates LEF1 activation in response to WNT1 and WNT3A, contributing to transcriptional regulation of Wnt target genes.
- Forms a ternary complex with PAK1 and MUSK that is critical for MUSK-dependent AChR clustering during neuromuscular junction formation.
- Localizes to the cell membrane, cytoplasm, cytosol, and cytoplasmic vesicles, enabling dynamic signaling from receptor complexes to intracellular effectors.
- Post-translationally modified by phosphorylation and ubiquitination (as indicated by keywords), which likely regulate its activity and stability.
- Alternative splicing generates multiple isoforms, potentially contributing to functional diversity in different tissues.
- Associated with developmental processes and human disease (e.g., dwarfism, based on keyword annotation).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 510-640) of human DVL1; therefore, the antibody may detect full-length DVL1 and any C-terminal fragments.
- For Western blot (WB), the expected molecular weight is approximately 75 kDa; alternative splicing may produce additional bands. Consider using appropriate positive controls such as HeLa or HEK293 cell lysates.
- In immunohistochemistry (IHC-P), antigen retrieval may be necessary; validate the staining pattern in tissues known to express DVL1, such as brain or muscle.
- For immunofluorescence (IF/ICC), optimize fixation and permeabilization conditions to preserve the native localization (cell membrane, cytoplasm, vesicles).
- ELISA applications may be optimized using recombinant DVL1 protein as a standard. Consider cross-reactivity when using mouse or rat samples.
