Biological Background: DUT Function and Localization
- DUT encodes deoxyuridine 5'-triphosphate nucleotidohydrolase, a mitochondrial and nuclear enzyme that hydrolyzes dUTP to dUMP and pyrophosphate (dUTP pyrophosphatase). Related references: PMID:17880943 PMID:8631816 PMID:8805593
- By cleaving dUTP, DUT prevents uracil misincorporation into DNA and supplies dUMP for de novo thymidylate biosynthesis, critical for DNA replication fidelity and genome integrity.
- DUT also acts as a negative regulator of peroxisome proliferator-activated receptor (PPAR) through its N-terminal domain, inhibiting PPAR dimerization with retinoid X receptor (By similarity).
- The protein is essential for embryonic development, as shown by functional studies (By similarity).
- Subcellular localization includes both nucleus and mitochondrion, with a transit peptide directing mitochondrial import.
- Tissue specificity: Isoform 3 is constitutively expressed, while isoform 2 expression is replication-dependent and its degradation coincides with cessation of DNA replication (at protein level).
- Keywords: Alternative splicing, Diabetes mellitus, Disease variant, Hydrolase, Magnesium, Nucleotide metabolism, Nucleus, Phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen sequence (amino acids 89–252) covers the catalytic domain of mature DUT; when using this antibody in western blotting, consider validating the detected band at approximately 27 kDa in the relevant sample system and account for possible isoform-specific migration.
- For immunohistochemistry (IHC‑P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization conditions as needed for your tissue or cell type; the nuclear and mitochondrial localization may require appropriate controls.
- In ELISA applications, include proper positive and negative controls and consider the polyclonal nature of the antibody when designing sensitive detection formats.
