Biological Background: DUSP22 Function and Localization
- DUSP22, also known as JKAP, JSP-1, LMW-DSP2, and MKP-x, is a dual specificity phosphatase that can dephosphorylate both phosphotyrosine and phosphoserine/threonine residues. Related references: PMID:24714587, PMID:38225265
- Activates the JNK signaling pathway, playing a role in cellular stress responses. Related references: PMID:11717427
- Inhibits T-cell receptor (TCR) signaling and T-cell mediated immune responses by dephosphorylating and inactivating the tyrosine kinase LCK, and by inducing proteasomal degradation of the E3 ubiquitin ligase UBR2. Related references: PMID:24714587, PMID:38225265
- May also modulate B-cell receptor signaling and B-cell function.
- Localizes primarily to the cytoplasm, consistent with its role in signal transduction cascades.
- Ubiquitously expressed, with highest levels in heart, placenta, lung, liver, kidney, and pancreas, indicating broad regulatory functions.
- Post-translationally modified by myristoylation and phosphorylation (as inferred from UniProt keywords), which may regulate its activity and localization.
- Belongs to the dual specificity protein phosphatase family, characterized by their ability to regulate MAP kinase signaling pathways.
Experimental Guidance and Technical Tips
- The immunogen used covers amino acids 1-184 of human DUSP22, suggesting the antibody may recognize the N-terminal region. For WB, consider a dilution range and validate with appropriate positive controls (e.g., DUSP22-expressing cell lines).
- As a polyclonal antibody, robust signal amplification is expected, but careful blocking and optimization may be needed to minimize background in IF/ICC.
- For ELISA, ensure optimal coating conditions and antigen presentation; consider testing with recombinant protein or tissue lysates from human samples.
- Verify cross-reactivity in your specific experimental system; the antibody is validated for human samples, and performance in other species is not guaranteed.
