Biological Background: DUSP14 Function and Localization
- DUSP14, also known as MKP6 or MKP-L, is a dual-specificity protein phosphatase that belongs to the MAP kinase phosphatase family.
- It functions as a negative regulator of mitogen-activated protein kinase (MAPK) signaling by dephosphorylating ERK, JNK, and p38 MAP kinases.
- In T-cell receptor (TCR) signaling, DUSP14 plays a negative role by dephosphorylating the MAP3K7 adapter TAB1, leading to its inactivation. Related references: PMID:24403530
- The protein is classified under hydrolase and protein phosphatase activities, and is a member of the reference proteome for human studies.
- Its molecular weight is approximately 22.3 kDa (calculated), consistent with the reported 22 kDa band in western blot applications.
Experimental Guidance and Technical Tips
- For western blot, a band at ~22 kDa is expected. Validate with appropriate positive and negative controls.
- For immunofluorescence/ICC, optimize fixation and permeabilization conditions to detect endogenous DUSP14, as its subcellular localization may be context-dependent.
- The polyclonal nature of the antibody may result in recognition of multiple epitopes, reducing the risk of signal loss due to post-translational modifications.
- Since the immunogen is nearly full-length, the antibody is likely to detect total DUSP14, but confirm specificity by siRNA knockdown or knockout models in the relevant sample system.
