Biological Background: DUSP13 Function and Localization
- Dual specificity protein phosphatase 13A (DUSP13A), also known as muscle-restricted DSP (MDSP), is a probable protein tyrosine phosphatase. Alternative splicing of the DUSP13 gene produces distinct isoforms including DUSP13A (Q6B8I1) and DUSP13B (Q9UII6).
- Exhibits phosphatase activity on synthetic substrates (PubMed:15252030, PubMed:29106959). Related references: PMID:15252030 PMID:29106959
- Plays a phosphatase activity-independent regulatory role in MAP3K5/ASK1-mediated apoptosis by preventing MAP3K5/ASK1 inhibition by AKT1.
- Shows no phosphatase activity on MAPK1/ERK2, MAPK8/JNK, MAPK14/p38, or MAP3K5/ASK1.
- Skeletal muscle-specific expression.
- Localizes to the cytoplasm.
- Computed molecular weight of DUSP13A canonical sequence is ~20.7 kDa; multiple isoforms range from 6 to 27 kDa due to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen covers full-length DUSP13A (aa 1–198); validate antibody specificity in relevant cell lines or tissues under appropriate experimental conditions.
- For Western blot, expect multiple bands corresponding to different isoforms; use molecular weight markers and specific controls to confirm target identity.
- In ELISA, optimize coating conditions and antibody dilutions for polyclonal detection; include appropriate positive and negative controls.
- Confirmed cross-reactivity with mouse and rat DUSP13 orthologs; verify reactivity in your species-specific samples using recommended lysates.
