Biological Background: DTX3L Function and Localization
- DTX3L (also known as BBAP, RNF143, or rhysin-2) is an E3 ubiquitin-protein ligase belonging to the deltex family, characterized by a RING-type zinc finger domain.
- It functions in DNA damage repair and interferon-mediated antiviral responses by forming a complex with PARP9 (PubMed:12670957, PubMed:19818714, PubMed:23230272).

PMID:12670957 PMID:19818714 PMID:23230272 - DTX3L monoubiquitinates multiple histones (H2A, H2B, H3, H4). Specifically, upon DNA damage, it catalyzes H4K91 monoubiquitination, which may license further H4 modifications like H4K20 methylation (PubMed:28525742, PubMed:19818714).
Related references:
PMID:28525742 PMID:19818714 - Together with PARP9 and PARP1, DTX3L-mediated ubiquitination facilitates the recruitment of 53BP1, RAP80, and BRCA1 to double-strand break sites (PubMed:23230272).
Related references:
PMID:23230272 - In antiviral defense, DTX3L ubiquitinates viral proteases from EMCV and HRV via Lys-48 linkages, targeting them for proteasomal degradation. It also promotes STAT1-driven transcription through H2B monoubiquitination and chromatin remodeling (PubMed:26479788).
Related references:
PMID:26479788 - Beyond its catalytic activity, DTX3L regulates endosomal trafficking by reducing ITCH-mediated ubiquitination of ESCRT-0 components HGS and STAM, thereby promoting CXCR4 sorting to lysosomes (PubMed:24790097).
Related references:
PMID:24790097 - Subcellular localization includes the cytoplasm, nucleus, and membranes of early endosomes and lysosomes. The protein is subject to acetylation and phosphorylation, and its expression is interferon-inducible.
Experimental Guidance and Technical Tips
- The immunogen comprises the C-terminal region (amino acids 601–740) of human DTX3L, which may contribute to the observed cross-reactivity with mouse and rat orthologs. For species not listed, verify sequence homology before use.
- In Western blotting, a band around 84 kDa is expected. Note that post-translational modifications or alternative splicing may produce additional bands; include appropriate controls.
- For immunofluorescence/immunocytochemistry, anticipate nuclear and cytoplasmic staining; endosomal or lysosomal localization may be detectable depending on cell type and stimulation conditions. Fixation and permeabilization should be optimized for the target compartment.
- This polyclonal antibody is suitable for ELISA and may be used to detect DTX3L in cell lysates or recombinant protein preparations. Titration in the specific assay system is recommended.
